The Challenge
Due to stability issues, initial attempts to synthesize a complex long-chain phospholipid were unsuccessful. This culminated in isomerization during the final step, converting the cis double bond to trans and resulting in a cis/trans mixture. Upon investigation, Exim-Indis discovered that the HPLC method being employed could not effectively separate this mixture, leading to an incorrect assumption that the sample was pure.
The Solution
Exim-Indis worked with our CDMO partner to develop an alternative synthetic approach. Using a chiral compound as the starting material, we were able to carefully circumvent acidic conditions and optimize reaction parameters like temperature and solvents. By carrying out quintessential esterification while protecting the hydroxyl group, we effectively eliminated the risk of hydrolysis. This new approach not only resolved the stability issues and eliminated the isomerization problem, but it also streamlined the process by reducing the number of steps involved.
In delivering a high-quality product that met our customer’s specifications. this experience provided the Exim-Indis team with valuable insights for future projects, reinforcing the importance of adaptability in synthetic processes.
